چکیده

Background and Purpose—The 3 gene products of the Na /Ca exchanger (NCX), viz, NCX1, NCX2, and NCX3, may play a pivotal role in the pathophysiology of brain ischemia. The aim of this study was to investigate the transductional and posttranslational mechanisms involved in the expression of these isoforms during oxygen and glucose deprivation and their role in endoplasmic reticulum Ca refilling in cortical neurons. Methods—NCX1, NCX2, and NCX3 transcript and protein expression was evaluated in primary cortical neurons by reverse transcriptase–polymerase chain reaction and Western blot. NCX currents (INCX) and cytosolic Ca 2

P 23: Apoptosis Following Cortical Spreading Depression in Juvenile Rats

Introduction: Repetitive cortical spreading depression (CSD) can lead to cell death in immature brain tissue. Caspases are involved in neuronal cell death in several CSD-related neurological disorders. Yet, whether repetitive CSD itself can induce caspase activation in adult or juvenile tissue remains unknown. Inducing repetitive CSD in somatosensory cortices of juvenile and adult rats in vivo,...

Plasma membrane-cytoskeleton-endoplasmic reticulum complexes in neurons and astrocytes.

The possibility that certain integral plasma membrane (PM) proteins involved in Ca(2+) homeostasis form junctional units with adjacent endoplasmic reticulum (ER) in neurons and glia was explored using immunoprecipitation and immunocytochemistry. Rat brain membranes were solubilized with the mild, non-ionic detergent, IGEPAL CA-630. Na(+)/Ca(2+) exchanger type 1 (NCX1), a key PM Ca(2+) transport...

C-terminal fragments of APP: Its neurotoxic mechanisms and involvement in gene transcription

Several lines of evidence suggest that some neurotoxicity in AD is due to proteolytic fragments of APP. In this study, we compared the potency of neurotoxicity induced by CT with that of A-beta neurotoxicity and our results showed that various CT peptide fragments (CTFs; CTF99, AICD, CTF31) caused neurotoxicity in cultured cells and primary cortical neurons, induced strong non-selective inward ...
